Exocytosis in Paramecium

In Paramecium caudate, trichinosis are IOWA secretors vesicles that are released from the cytoplasm. We first observed how the presence of Alicia blue dye triggers normal secretion of trichinosis, and we then tested four different hypotheses in order to determine how the presence of Ca++ ions in the extracurricular and intracellular environment affects the secretion of trichinosis.Our first our hypothesis tested whether extracurricular calcium is required for the secretion of trichinosis by reading cells with EST., an agent that blocks calcium ions; our second tested whether increased calcium will counter the inhibitory effects of EST.; our third tested whether adding different ion, like Magic, can also counter the inhibitory effects of EST.; and our fourth hypothesis tested whether the release of intracellular calcium by adding caffeine or oenophile affects exoticism.Our results demonstrate that exoticism of tricycles is dependent on the presence of calcium ions in both the intracel lular and extracurricular environments. Introduction: Exoticism in eukaryotic cells is the process in which macromolecular material is released from the cell into the extracurricular environment; one example of exoticism is the secretion of trichinosis in paramecium. Trichinosis are secreted from the cell as a defense mechanism when certain stimuli are introduced into the extracurricular environment.The secretion of trichinosis is controlled by the presence of Ca*+ ions within the plasma membrane and in the extracurricular environment. The purpose of our experiment was to observe the extent to which exoticism is pendant on the presence of extracurricular and intracellular calcium ions, and to determine if other extracurricular ions can have the same effect as calcium. The cells were first treated with 0. 03% Alicia blue dye, which triggers normal exoticism. Cells were then treated with EST., an agent that inhibits calcium, in order to observe how triptychs secretion is affected when calcium ions are sequestered.Additional calcium ions were added to the extracurricular environment to determine if the effects f EST. could be countered by increasing calcium. The cells were then exposed to a different ion, Magic, to observe if a different ion could have the same promoting effect in the extracurricular environment as calcium. Finally, the effect of intracellular calcium was tested by treating cells with a calcium oenophile (A23187) and caffeine, which both stimulate the processing of calcium into the cytoplasm of a cell. Our class will be going outside to try to help us understand Paleolithic times. We will experience the world as Paleolithic people did – bringing only what we can carry, having no permanent shelters, creating our own art from the materials around us and having to figure out the best place to stay. For this activity, we will assume that you are traveling in a small family group that Is part of a larger group of 31. You can be with either two other people or work with a partner, representing the core family group. It was typical of Paleolithic times that related core family groups would join together o form a larger group.This â€Å"family† would work together to gather food and protect each other. The group was usually between 15 and 30 people, though smaller and larger groups existed. As they traveled, different family groups would meet to exchange goods that they had made and they likely shared Information about the local environment. Young people likely moved between groups in order to share expertise (a group with several artists might exchange an artist for a skilled hunter) or to meet the needs of both groups (a group with extra males may send some to another group in exchange for goods or services).Over time, the larger group size allowed for some specialized roles to develop. The majority of people in the group would supply food for everyone, giving some people free time. For instance, a religious leader, artist or expert craftsman might supply the group's spiritual or technological needs rather than hunting or parenting. Women could take on a leadership role in the group. Women who gathered plants and hunted for small animals in many cases supplied most of the food for the group. In their role as mothers, they were protected from the most dangerous tasks, such as hunting or participating In battles with enemies.Instead, these types of tasks would be left to the â€Å"extra† males (the young and unattached men who were not yet In re ligious, artistic or craftsmen roles). An Evaluation of the Myers Briggs Type Indicator The Myers-Briggs Type Indicator (MBTI) is a psychological test that was originally designed in the early 1940s by a mother-daughter team of Isabel Myers and Katherine Briggs. Myers and Briggs developed their test in response to the calamity of World War II, because they believed the war was caused by the countries involved failing to understand each other’s differences (Coe, 1992). The MBTI is designed to measure personality, and the basis for this test comes from the work of personality types as described by Carl Jung’s theory. All the information provided in this evaluation comes from form M of the MBTI. Purpose, Design and Format of the MBTI The purpose of the MBTI is to classify individuals on each of the four levels as identified in Jung’s theory, and then provide them with a description of their personality as a result of their test (Fleenor, 2001). On each of the four levels to identify a person there are two options on each level making a total of eight different possibilities that can be used to describe someone’s personality. Those levels are: Extraversion v. Introversion, Sensing v. Intuition, Thinking v. Feeling, and Judging v. Perceiving. As defined for the MBTI Extraverts enjoy communication with others and thrive off that contact; while Introverts tend to not find any pleasure in their relationships and prefer to be alone. Sensors use their five senses to gather information about a scenario and see the present for what it is; in contrast people scoring high in Intuition like to look toward the future and the possibilities that could exist. Finally, Judgers use the information available and come to a decision quickly; whereas Perceivers tend to be procrastinators and are always waiting for more facts before deciding (Coe, 1992). It is important to note that while the MBTI places test takers in a category of having one trait or the other that people do display characteristics of all eight, but tend to display one aspect on each level more prominently. The degree to which a person varies on a particular attributed is explained upon completion of the test. The questions on the MBTI are also designed in such a way that it is impossible to fall in the exact middle of a particular trait, everyone will vary slightly one direction or another (Fleenor, 2001). This test is designed for the general population ages 14 and older, and there is not a specific group who is not eligible to take the MBTI. This is considered a weakness by many and will be covered later. There are 93 multiple choice items on the MBTI, and the questions are written at a seventh grade reading level to ensure comprehension of the questions (Fleenor, 2001). Each question is designed so that one question is measuring just one type of personality level for simple and accurate scoring. In defining the norms for this test a group of 3009 people were administered the MBTI all were adults from the United States and were 18 years and older. Th e specific demographics of the normative sample are not given, but the sample was lopsided towards females and Caucasians (Fleenor, 2001).

Steve Gregg wants the bike to be utilized for his department as he is handling the most important project in the plant which will necessitate Debbie to buy a new bike for Ronnie's inspection. At the start of the case, it was stated that Debbie's boss is "not an easy person to work for" since he expects his employees to have a "take charge attitude and avoid bothering him with trivial issues." Taking this into account, Debbie should try to resolve the problem at hand first before resorting to his immediate superior.
